The US National Organization for Rate Disorders (NORD) has more than 1,200 diseases in their list while National Institute of Health (NIH)’s office for rare diseases published a list of more than 6,000 disease disorders ranging from Aagenaes syndrome to Zuska’s Disease. In Europe, orphan diseases are those that have a prevalence of less than 5 per 10,000 of the population. According to European Organization for Rare Diseases (EURORDIS), there are about 6,000 to 8,000 orphan diseases that exist in Europe. In Japan, orphan diseases are those that have a prevalence of less then 4 per 10,000 (Orphan Drug Act 1993).

The regulatory bodies play a crucial role in defining the orphan diseases as well as providing incentives to manufacturers who are involved in development of orphan drugs. Before the approval of orphan drugs, every molecule is designated as an orphan drug and will enter clinical trials. It is approved as an orphan drug only after the molecule clears the various phases in a clinical trial.
